摘要:

代谢组学是定量研究生物体内源性代谢物整体及其变化规律的科学,是系统生物学的一个重要组成部分. 通过代谢组分析,可以较为全面地认识给定生物系统受内因或外因的影响后其生物化学过程变化的规律. 目前,代谢组分析检测技术主要包括核磁共振波谱技术及色谱-质谱联用技术. 代谢组数据分析和挖掘技术主要包括非监督与监督的多变量分析模式识别方法. 目前已被广泛应用于病理生理、基因功能、药物毒理和环境毒理等领域. 该文简要综述了代谢组NMR分析中的实验设计、NMR谱的获取、模式识别技术和代谢组学在药物毒理评价中的应用. 

Abstract:

Metabonomics studies the variations of levels of endogenous metabolites in response to physiological rhythms and external stimuli, providing molecular mechanistic explanations to the effects of pathology, toxins or specific environmental conditions. It has become an important tool in systems biology. With metabonomic approaches, mass spectrometry or nuclear magnetic resonance (NMR) spectroscopy data acquired from biological samples are analyzed with pattern recognition techniques. These approaches have been successfully applied in many research areas, including biomedicine and drug toxicology. In this review, we focused on experimental design, assessment of NMR spectroscopy, pattern recognition techniques and applications of NMR-based metabonomic approaches in studying drug toxicology.
