摘要: 用二维核磁共振的方法确定二锌胰岛素质子核磁共振谱中的甲基,亚甲基基团共振峰。结合二锌胰岛素x-射线晶体结构所提供的信息,通过欧弗豪斯效应二维偶极相关谱(NOEsY)已确定10个异亮氨酸,亮氨酸,缬氨酸,苏氨酸甲基基团共振峰。由偶合相关二维谱(COSY)交叉峰的分析可确定13个其它基团的共振峰的归属。在pH=11.0时,二锌胰岛素以单体形式出现。自旋扩散不是一个严重问题。

Abstract: Two-Dimensional NMR methods were used to assign methyl and methylene group resonances in the NMR spectra of 2-Zn insulin. 10 methyl group of Ile, Leu, Val,Thr resonances are specifically assigned using nuclear overhauser effect (NOESY) spectra of insulin in combination with the information of X-ray structure.
Assignment of other group resonances were based on the analysis of Two-Dimensional spectra of the J-Correlated (COSY).
Insulin exists in the monomeric state at pH 11.0, where spin diffusion is not a serious problem.
