采用WET溶剂抑制方法对mHCN1孔隙多肽1H谱线进行归属

摘要:

孔隙部分是离子通道选择性控制的关键部分. 与K+道相似,HCN离子通道孔隙区域包括GYG特征序列. 但除了K+外,Na+也能选择性通过HCN离子通道. 对mHCN1孔隙多肽 的构象研究将有助于对HCN离子通道选择性的理解. 本文利用新开发的WET溶剂抑制方法并综 合运用wet-TOCSY和wet-NOESY NMR方法对mHCN1孔隙多肽(95%水+5%重水)进行了1H谱 线归属. 

Abstract:

Like K+ channel, the pore region of HCN channel includes GYG characteristic se quence. But besides K+, Na+ can selectively overpass HCN channel. The conf ormation study of mHCN1 pore region peptide helps to understand the selectivity  of HCN channel. In this article, new developed WET sequence was used to suppress  solvent peak. With the comprehensive use of wet-TOCSY and wet-NOESY NMR exper iments, the 1H resonance of mHCN1 pore region peptide was assigned.(95%H2O+5%D2O).
